No, marrying is not a requirement to be a naturalized Japanese citizen.
Though it is the easiest way to be one.
I know several Catholic priests in Japan, who used to be foreigners but
became Japanese citizens with Japanese names. They are, of course, all
singles.
You need to speak Japanese, have an occupation, has been living in Japan
for an extended years and have a few Japanese sponsors who could officially
give an assurance of the person's good behaviors, or something like that.
